TAE buffer is widely used as a nucleic acid electrophoresis buffer, mainly for DNA agarose gel electrophoresis in biology. When performing electrophoresis of segments larger than 13 kb, using TAE buffer achieves a better separation effect. It is also easy to use the TAE buffer system for electrophoresis when recovering DNA fragments. The disadvantage of TAE is its small buffer capacity, which cannot be used for long-term electrophoresis (such as overnight).

1. Weigh 242g of Tris and 37.2g of Na2EDTA.2H2O in a 1L beaker;
2. Add approximately 800ml of deionized water to the beaker and stir thoroughly until uniform;
3. Add 57.1ml of glacial acetic acid and dissolve thoroughly;
4. Add deionized water to a constant volume of 1L and store at room temperature.
